1. First, remove the inner bucket, then add an appropriate amount of water to the outer pot, ensuring that the water level is flush with the triangular rack.

2. Place the items back into the sterilization bucket and load them carefully, ensuring they are not packed too tightly to allow free steam circulation and maintain effective sterilization. Neither the neck of the Erlenmeyer flask nor the opening of the test tube should come into contact with the bucket walls, as condensation could drip onto the wrapping paper at the bag opening and penetrate the cotton plug.

3. Install the lid and insert the vent hose on the lid into the vent groove of the inner bucket. Then, tighten the two opposing bolts simultaneously in a symmetrical, pair-by-pair manner to ensure even tension and prevent air leakage.

4. Heat the autoclave using an electric heater or a gas burner, and simultaneously open the exhaust valve to bring the water to a boil, thereby expelling the cold air from the chamber. Once all the cold air has been completely vented, close the exhaust valve and allow the temperature inside the chamber to rise gradually as the steam pressure increases. When the chamber pressure reaches the desired level, reduce the heat source and maintain that pressure for the required duration. In this experiment, the settings are 1.05 kg/cm², 121.3°C, and 20 minutes. 5. After the prescribed time has elapsed, disconnect the power supply or turn off the gas, and allow the chamber temperature to drop naturally. When the pressure indicated by the gauge falls to zero, open the exhaust valve, loosen the bolts, lift the lid, and remove the items. If the pressure has not yet dropped to zero when the exhaust valve is opened, the sudden release of pressure inside the chamber will cause the culture medium to be forced out of the flask or test tube due to the resulting pressure imbalance between the inside and outside of the container, leading to contamination of the cotton plugs with the medium.

6. Place the retrieved culture medium in a 37°C incubator for 24 hours; if no contaminating microorganisms are observed upon inspection, it is ready for use.

However, there are a few points to keep in mind: 1. The pot must contain sufficient water. 2. The pot must be completely airtight.

3. After use, be sure to allow it to reach the appropriate temperature before opening.
