It is essential to insert the vent hose on the lid into the vent groove of the inner bucket, and then tighten the two opposing bolts simultaneously in a symmetrical, pair-by-pair manner, ensuring there are no leaks.

　　Once the required time has elapsed, be sure to disconnect the power supply or shut off the gas valve to allow the temperature inside the autoclave to drop naturally. Only when the pressure indicated by the gauge has fallen to zero should you open the exhaust valve, loosen the bolts, lift the lid, and carefully remove the items. Of course, if you notice that the pressure gauge has not yet dropped to zero, you must not open the exhaust valve under any circumstances. Opening the valve would cause a sudden drop in pressure inside the chamber, leading to an imbalance between the internal and external pressures. This could force the culture medium out of the flask through the opening, contaminating the cotton plug and resulting in contamination of the medium. In such a case, the entire procedure would be considered a failure.
